Menopausal hot flashes and the default mode network

Objective
To test whether more physiologically assessed hot flashes were associated with more connectivity in the default mode network (DMN), the network of brain regions active during rest. We particularly focus on DMN networks supporting the hippocampus as this region is rich in estrogen (E) receptors (ER) and has previously been linked to hot flashes.
Design
Women underwent 24 hours of physiologic and diary hot flash monitoring, functional magnetic resonance imaging (MRI), 72 hours of sleep actigraphy monitoring, a blood draw, questionnaires, and physical measures.
Setting
University medical center.
Patient(s)
Twenty midlife women aged 40–60 years who had their uterus and both ovaries and were not taking hormone therapy (HT).

Main Outcome Measure(s)
The DMN functional connectivity.
Result(s)
Controlling for age, race, and education, more physiologically-monitored hot flashes were associated with greater DMN connectivity (beta, B [SE] = 0.004 [0.002]), particularly hippocampal DMN connectivity (B [SE] = 0.005 [0.002]). Findings were most pronounced for sleep physiologic hot flashes (with hippocampal DMN, B [SE] = 0.02 [0.007]). Associations also persisted controlling for sleep, depressive symptoms, and serum E2 concentrations.
Conclusion(s)
More physiologically-monitored hot flashes were associated with more DMN connectivity, particularly networks supporting the hippocampus. Findings were most pronounced for sleep hot flashes. Findings underscore the importance of continued investigation of the central nervous system in efforts to understand this classic menopausal phenomenon.

Neurobiological Factors as Predictors of Prisoners’ Response to a Cognitive Skills Training

Purpose
The current study investigates the predictive value of neurobiological factors in relation to detainees’ treatment outcome, in order to better understand why some individuals respond favorably to treatment while others do not. It was hypothesized that low levels of heart rate activity are associated with poor treatment outcome and that weak neurocognitive functioning is predictive of more benefit from therapy.
Methods
Background characteristics, behavioral measures, neurocognitive functioning and heart rate activity of 121 male detainees selected for cognitive skills training were assessed. Outcome measures included program completion, evaluations by trainers and ward staff, and detainees’ self-reported motivation and treatment evaluation.
Results
Concentration performance, a neurocognitive skill, significantly predicted treatment dropout over and above background and behavioral measures, including self-reported motivation. In addition, high selfreported ‘meanness’, a psychopathic feature, was associated with low treatment motivation and an expectation bias seemed to be present among highly motivated detainees. These results did not confirm the hypotheses.
Conclusions
Offenders who are characterized by a decreased concentration performance, low motivation and increased meanness, are less likely to benefit from treatment. The results have the potential to improve the current treatment assessment procedures in order to reduce dropout rates and, eventually, recidivism rates.

Lab And/Or Field? Measuring Personality Processes and Their Social Consequences

How can researchers study personality processes and their social consequences? In our methodology overview, we first introduce ambulatory assessment methods, which repeatedly measure experiences, physiology and behaviour in people’s daily lives based on real?time assessments of self?reports, physiological activity and behavioural observations. Then, we describe methods suitable for assessing personality processes in laboratory settings: self?reports on interpersonal perception, physiological measurements and behavioural observation. We discuss the combination of field and laboratory assessment methods based on their respective strengths and limitations and then highlight ethical issues surrounding the use of these methods. Finally, we propose future avenues for how developments in mobile technology can be used to advance personality research. The increasing availability and the decreasing costs of smartphones, wearable sensors and Internet connectivity offer unique potentials for further understanding the processes underlying how personality exerts broad and important social consequences. Attachment and physiological reactivity to infant crying in young adulthood: Dissociation between experiential and physiological arousal in insecure adoptees

The associations between attachment representations of adopted young adults and their experiential and physiological arousal to infant crying were examined. Attachment representations were assessed with the Attachment Script Assessment (ASA), and the young adults listened to infant cries, during which ratings of cry perception were collected and physiological reactivity was measured. Secure adoptees showed a well-integrated response to infant distress: heart-rate increases and respiratory sinus arrhythmia (RSA) withdrawal were coupled with heightened perception of urgency in these individuals. In insecure adoptees RSA withdrawal was absent, and a combination of lowered perceived urgency and heightened sympathetic arousal was found, reflecting a deactivating style of emotional reactivity. Overall, our findings support the idea that internal working models of attachment explain individual differences in the way attachment-related information is processed.

Testing Van Gool’s Hypothesis: A Method to Predict Side Effects of Cholinesterase Inhibitors in Patients with Cellular Degenerative and Vascular Dementia

This study investigates a method to predict medical outcome of cholinesterase inhibitors in patients with Alzheimer’s disease (AD) and vascular dementia (VaD). Van Gool predicts that patients with cholinergic deficit symptoms will benefit from treatment whereas patients without will experience side effects because of overstimulation of the cholinergic system. We predicted that AD and VaD patients with a longer RT experience fewer side effects than patients with a faster response and that VaD patients have a longer RT than AD patients. A number of 71 patients with AD or VaD diagnosis were included. A sustained attention task was administered, as well as the MMSE and a questionnaire about side effects. Results indicated that VaD patients with a longer RT reported fewer side effects. Furthermore, patients with VaD had a longer RT than patients with AD. MMSE was negatively correlated with RT in the VaD group. Thus, the performance on the attention task seems associated with suffering from side effects and thus tends to predict medical outcome in VaD, but not in AD. Perhaps this attention task was not sensitive enough to measure cholinergic deficit symptoms in AD patients. Furthermore, different doses of medication might confound the effect for the AD group.

Using basic neurobiological measures in criminological research

Research on the relationship between neurobiological factors and antisocial behavior has grown exponentially in recent decades. As a result, criminal behavior has been related to impairments in different biological systems, such as genetics, hormones and brain functioning. The development of innovative techniques, for example brain imaging techniques and physiological measurements, can partially explain the increase in neurobiological studies on criminal behavior. Despite a recent zeitgeist change, which has led to a greater acceptance of neurobiology as an additional approach for the study of criminal behavior, neurobiological measurements dot not yet play a more significant role in criminological research and practice. This article aims to familiarize crime researchers and other interested readers with two important neurobiological measures, namely neuropsychological assessment and resting heart rate measurement, and with how these measures can play an important role in criminological research. It will argue that neuroscientific approaches might benefit the field of criminology in several important ways, from assessing the empirical validity of criminological theories to improving the effectiveness of correctional intervention programs. Furthermore, this article will provide insight into how to use these measures in research, to guide interested readers towards their application in their own studies.

Pain in dementia: prevalence and associated factors: protocol of a multidisciplinary study

Pain is a common problem in people with dementia, however the exact prevalence of pain in dementia subtypes, e.g. Alzheimer’s Disease (AD), Vascular Dementia (VaD), Frontotemporal Dementia (FTD) and dementia with Lewy Bodies (DLB), is unknown, as is the relation between pain and the different subtypes of dementia. In this study, the prevalence of pain in people with dementia will be investigated per dementia subtype and the relationship between the various subtypes of dementia and the presence of specific types of pain (i.e. musculoskeletal pain, neuropathic pain and orofacial pain) will be examined. Secondly, associations between various types of pain, cognitive functioning, neuropsychiatric symptoms and quality of life in people with dementia will be examined. A third purpose is to study the value of the assessment of autonomic responses in assessing pain in people with dementia. Finally, the effect of feedback to the attending physician on the presence of pain, based on examination by investigators with backgrounds in neuropsychology, geriatric dentistry and elderly care medicine, will be evaluated.

Progenitor cells are mobilized by acute psychological stress but not beta-adrenergic receptor agonist infusion

Objectives
Stimuli that activate the sympathetic nervous system, such as acute psychological stress, rapidly invoke a robust mobilization of lymphocytes into the circulation. Experimental animal studies suggest that bone marrow-derived progenitor cells (PCs) also mobilize in response to sympathetic stimulation. Here we tested the effects of acute psychological stress and brief pharmacological β-adrenergic (βAR) stimulation on peripheral PC numbers in humans.
Methods
In two studies, we investigated PC mobilization in response to an acute speech task (n=26) and βAR-agonist (isoproterenol) infusion (n=20). A subset of 8 participants also underwent the infusion protocol with concomitant administration of the βAR-antagonist propranolol. Flow cytometry was used to enumerate lymphocyte subsets, total progenitor cells, total haematopoietic stem cells (HSC), early HSC (multi-lineage potential), late HSC (lineage committed), and endothelial PCs (EPCs).
Results
Both psychological stress and βAR-agonist infusion caused the expected mobilization of total monocytes and lymphocytes and CD8+ T lymphocytes. Psychological stress also induced a modest, but significant, increase in total PCs, HSCs, and EPC numbers in peripheral blood. However, infusion of a βAR-agonist did not result in a significant change in circulating PCs.
Conclusion
PCs are rapidly mobilized by psychological stress via mechanisms independent of βAR-stimulation, although the findings do not exclude βAR-stimulation as a possible cofactor. Considering the clinical and physiological relevance, further research into the mechanisms involved in stress-induced PC mobilization seems warranted.

Heritability and Temporal Stability of Ambulatory Autonomic Stress Reactivity in Unstructured 24-Hour Recordings

Objective 
Measurements of ambulatory autonomic reactivity can help with our understanding of the long-term health consequences of exposure to psychosocial stress in real-life settings.
Methods 
In this study, unstructured 24-hour ambulatory recordings of cardiac parasympathetic and sympathetic control were obtained in 1288 twins and siblings, spanning both work time and leisure time. These data were used to define two ambulatory baseline (sleep, leisure) and four stress conditions (wake, work, work_sitting, work_peak) from which six ambulatory stress reactivity measures were derived. The use of twin families allowed for estimation of heritability and testing for the amplification of existing or emergence of new genetic variance during stress compared with baseline conditions.
Results 
Temporal stability of ambulatory reactivity was assessed in 62 participants and was moderate to high over a 3-year period (0.36 < r < 0.91). Depending on the definition of ambulatory reactivity used, significant heritability was found, ranging from 29% to 40% for heart rate, 34% to 47% for cardiac parasympathetic control (indexed as respiratory sinus arrhythmia), and 10% to 19% for cardiac sympathetic control (indexed as the preejection period). Heritability of ambulatory reactivity was largely due to newly emerging genetic variance during stress compared with periods of rest. Interestingly, reactivity to short standardized stressors was poorly correlated with the ambulatory reactivity measures implying poor laboratory–real-life correspondence. Conclusions  Ambulatory autonomic reactivity extracted from an unstructured real-life setting shows reliable, stable, and heritable individual differences. Real-life situations uncover a new and different genetic variation compared with that seen in resting baseline conditions, including sleep.

Cardiac Autonomic Nervous System Activation and Metabolic Profile in Young Children: The ABCD Study

Background In adults, increased sympathetic and decreased parasympathetic nervous system activity are associated with a less favorable metabolic profile. Whether this is already determined at early age is unknown. Therefore, we aimed to assess the association between autonomic nervous system activation and metabolic profile and its components in children at age of 5–6 years. Methods Cross-sectional data from an apparently healthy population (within the ABCD study) were collected at age 5–6 years in 1540 children. Heart rate (HR), respiratory sinus arrhythmia (RSA; parasympathetic activity) and pre-ejection period (PEP; sympathetic activity) were assessed during rest. Metabolic components were waist-height ratio (WHtR), systolic blood pressure (SBP), fasting triglycerides, glucose and HDL-cholesterol. Individual components, as well as a cumulative metabolic score, were analyzed. Results In analysis adjusted for child’s physical activity, sleep, anxiety score and other potential confounders, increased HR and decreased RSA were associated with higher WHtR (P< 0.01), higher SBP (p<0.001) and a higher cumulative metabolic score (HR: p < 0.001; RSA: p < 0.01). Lower PEP was only associated with higher SBP (p <0.05). Of all children, 5.6% had 3 or more (out of 5) adverse metabolic components; only higher HR was associated with this risk (per 10 bpm increase: OR = 1.56; p < 0.001). Conclusions This study shows that decreased parasympathetic activity is associated with central adiposity and higher SBP, indicative of increased metabolic risk, already at age 5–6 years.
